Clone REA765 recognizes the human CD137 antigen, a 30 kDa glycoprotein of the tumor necrosis factor (TNF) receptor superfamily, a group of cytokines that can cause cell death (apoptosis). CD137 is mainly expressed on activated CD4+ and CD8+ T cells, activated B cells, and natural killer cells, but can also be found on resting monocytes and dendritic cells.

CD137 is a 39 kD transmembrane protein also known as 4-1BB. It is expressed on activated T cells. CD137 is a type I membrane protein and a member of the tumor necrosis factor receptor superfamily. CD137 appears to be important for T cell proliferation and survival, and induces monocyte activation through its interaction with 4-1BB ligand. Agonist monoclonal antibodies (mAb) to the immune costimulatory molecule CD137, also known as 4-1BB, are presently in clinical trials for cancer treatment on the basis of their costimulatory effects on primed T cells and perhaps other cells of the immune system. Here we provide evidence that CD137 is selectively expressed on the surface of tumor endothelial cells. Hypoxia upregulated CD137 on CD137-PE recognizes the CD137 (4-1BB) antigen, a 30 kDa glycoprotein of the tumor necrosis factor (TNF) receptor superfamily.
